The Dubois family felt driven to move the research forward and help to get it to the finish line! Working hard they found the perfect bakery to partner with to make a big difference! PBD Canada is hosting a $500 Cash Prize Lottery! Tickets are $2 each and can be purchased through PBD Canada team members. Ticket sales close July 31st and the winner will be announced August 1st, 2018 on the PBD Canada Facebook page.

The money donated to Dr. Braverman's lab will be used to advance several research projects that are underway, for example:
- Research of drugs using the skin cells of patients with mutations in different PEX genes: We test different drugs or molecules to find which ones improve the function of peroxisomes, or in other words which ones allow the peroxisomes to work well enough for the cells to regain their health. We then move forward with interesting drugs to test them in our mouse model. - Gene editing in cell cultures: We use the latest CRISPR / Cas9 methods to introduce specific mutations into the PEX genes of cells in Petri boxes. This will allow us to understand the effects of specific mutations on peroxisome functions. We will soon be working with these cells in culture to correct the mutations found in patients, and we want to test whether we can correct the defective peroxisomes in this way. - Mouse model: Mouse model: Our mouse model has two copies of the most common mutation in the ZSD, the PEX1 G843D. We use these models to first understand how the disease evolves in the mouse (is it like in humans? What can we learn about the course of the disease?), and then to test our interesting candidate drugs. We can study specific organs and systems in the mice, their longevity, their growth and others, after a treatment compared to a placebo. One of our current projects is to test a gene replacement therapy in the retina of the mice to see if it improves their vision. In the future, we would like to test gene therapy in other organs and systems in the mouse and see if it improves the course of their disease. - Natural history of patients with PBD: We collect medical records, we transfer the data into a database to do large-scale analyses. Our goal is to characterize the natural progression of clinical symptoms in people with PBD in order to improve management guidelines and to determine endpoints for future clinical trials. The information collected in this study is crucial to move forward to clinical trials. All clinical trials require a well-described natural history prior to start the trial. - We want to measure the levels of PEX1-G843D-modified protein in patients skin cells, to determine if there is a correlation between the level of this modified protein and the severity of the disease in patients. - We want to find biomarkers to measure in patients that would be more reliable and less variable to evaluate their response to therapies (proteomics, metabolomics, lipidomics). - We want to find modifying elements in patients that would be predictive of specific clinical symptoms, such as leukodysrophies. Could specific symptoms be influenced by a genetic peculiarity, by the environment, by the diet, or by some drugs? The question remains unanswered but we hope to better understand why symptoms and their severity vary in each person affected with PBD. |
